Introduction:
Planarian(flatwormDugesiajaponica)isoneofaseriesofanimalspeciesthathavetheabilitytore-growtheirbodypartswhendamaged.Whenitsbodyiselongated,itissoft,leaf-shapedandciliated.Ithasspade-shapedheadwithtwoeyesandtentacleswhilethetailispointed.Itsmouthisevenontheventral.Whenitcomestoitsinnerstructures,ithasnobodycavity(coelom),soitspharynxmayendsinan
intestine.Itusuallyis3to15mmlong,butitcanalsogrowtomorethan30cm.Itmovesbyswimminglikeslugs,tendingtoseekfoodwhichincludesprotozoans,tinysnailsandwormsatnightascarnivorouscreature.
Planariansarehermaphroditesandtheirreproductiveorgansstarttodevelopinearlyautumn,buttheycannotinbreed.Mostoftheseindividualsareabletofullydevelop,whilesomemaybeabnormal.Andbecauseofplanarians’remarkableabilitytoregeneratelostparts,theyareoftenusedinbiologyexperiments.
Inthisexperiment,wewillhaveanopportunitytoobserveandrecognizesomeofthebasicfeaturesofplanarians.Wewillbetaughttousescientificapproachtohypothesizehowplanariansrespondtofragmentation.Theplanarianwillbeworkedasmodel.
Materialsandmethods:
Materials:
Stereoscope(Nikon),Handlenses,Petridishwithfrozenwater,Knife,Tweezers,Droppers,Filterpapers,Planarians.
Methods:
PartⅠ:
Observetheplanarian.
⑴Usehandlensestohaveacloseobservationonplanarian,andrecordwhatareseenintableT-1.⑵Identifythefollowingfeaturesandmakenotesaccordingly:
color,shape,movement,length,
symmetry,visiblebodyparts,responsestolight,foodandflippingtheanimalupsidedown.⑶Summarizewhatwehavefoundout,andtrytofindconnectionsbetweendifferentaspectsof
planarian’sbiology.
PartⅡ:
ExploringregenerationinPlanariansbycuttingaPlanarian.⑴Labelapetridishwithmarkpen.
⑵PlaceaPlanarianonwetfilterpaperplacedontopofbedoffrozenwaterinapetridish,sothatitcanberelativelystill.
⑶BisectthePlanarianintotwopieces,like⑷UseadroppertodislodgethePlanarianoffthefilterpaperandintothelabeledpetridish.Fill
thepetridishabouthalffullwithwater.
PartⅢ:
ObserveregenerationswhichcomefromtrisectionofPlanarianovertime,basedonthesamples
providedbyteachers.
⑴CutaPlanarianhowwewantto,placethepartsintothelabeledpetridish,fillitabouthalf
fullwithwater.
⑵ObserveatdifferenttimePoints(2hours,1day,2days,3days,7days),andmakenotesabout
thechangesincolorandtheprogressionoftheregeneratingpartsintableT-2.

TheshapeofPlanarianbenefitstoitslivinginwater,suchasaspade-shapedhead.Andwhenfeeditmeat,
itwillapproachtotheobjectiveslowly;thenifnothingthreatenit,itwillreachoutitsmouthparttothefood.
Whenitmoves,itwilltryitsbesttoavoidlightsource.

Afterbeingcuttotwoparts,theycanbothstillmovesinpetridish.Relatively,thepartofheadismoreactivethanthepartoftail.ThefrozenwaterdoesmakethePlanarianremainrelativelystill,sothatitiseasiertocut.[ThedevelopmentofthesetwopartswillbegiveninTableT-2].

Intheteacher’ssamplesaretrisectionofPlanarian,thetimeis4.30,5.4,5.5,5.6,5.7respectively,whentheyareobservedon5.7.
BesidesthesamplefromPartⅡ,inoursamples,thePlanarianiscutfromheadtotail(bisect),likeTableT-2(AsshownintheseriesofFigure3)

Planarianisalowformoflife,thestructureisrelativelysimple.Soitisabletoregeneratebycelldifferentiation.Butcellsofhumankindhavebeenhighlydifferentiated,thestructureisverycomplex,regeneratingmaymakedaughtercellswrongwhichleadstodeath.
Themorehighlythecreaturedifferentiates,theharderittendstobeabletoregenerate.Forexample,highformoflife’ssomaticcellsarealmostallhighlydifferentiated,therearehardlyothercellstomakeupthelostpartoftheorgan.
